The film Chal Mere Bhai tells the tale of two brothers, Vikram ‘Vicky’ Oberoi (Sanjay Dutt) and Prem Oberoi (Salman Khan), and how a young woman named Sapna (Karisma Kapoor) completely changes their lives. Vicky is a corporate mogul who oversees the vast business empire of his family. Sapna submits an application to be his secretary. Although Sapna doesn't have the necessary expertise to be Vicky's secretary—she is an orphan who recently migrated to Bombay from Pune and is now living with her uncle and aunt—Vicky's father, Balraj Oberoi (Dalip Tahil), is inspired by Sapna's devotion and recruits her.Prem wants to be an actor, much to Balraj's dismay because Balraj wants Prem to work for the family business. Vicky and Prem's grandmother (Sushma Seth) do, however, encourage his decision to pursue acting. He collaborates with theatrical director Shaktik Kapoor, Sapna's uncle.Vicky is always annoyed by Sapna's clumsy manner and lack of skill at work. But Sapna's quick thinking saves Vicky's life when he is attacked after work. Vicky's relatives extend an invitation to her and her family for a little getaway at their farm. Sapna won over Vicky's family, who grew to love her. At this time, Prem and Sapna also fell in love.

But before Prem can tell his family that he loves Sapna, he learns that Vicky has accepted his grandmother's proposal to marry Sapna. Prem is reluctant to hurt his brother's feelings. For a very long period, Vicky has been mourning the loss of his deceased girlfriend Piya (Sonali Bendre). Vicky has finally made up her mind to go on, and Prem doesn't want to come in the way of that. Prem thus misrepresents his relationship with Sapna by saying that they were just having fun. A shattered Sapna caves in to her aunt and uncle's persuasion and consents to marry Vicky. Before the wedding, Vicky learns the truth, and she confronts them. At the conclusion, Sapna and Prem are reconciled. In the final scene, it is implied that Vicky rediscovers love when Pooja (Twinkle Khanna), a fresh girl, joins his office.
