In an orphanage in Ooty run by businessman Jamnadas, there are six orphans: Gopal, Madhav, Lucky, Laxman, and Laxman 2. Lucky is deaf, Laxman struggles with his speech, and Gopal has a fear of ghosts. The lads adopt a baby girl they find and give her the name Khushi. They are concerned for her and are aware of Anna the librarian's propensity for seeing ghosts and spirits. A battle breaks out between the boys one day when Madhav scares and plays tricks on the timid Gopal in an unoccupied bungalow. After receiving a penalty, Gopal and Laxman decide to leave the orphanage. Soon after, Madhav, Lucky, and Laxman follow suit. Later, the devoted Colonel Chauhan and his wife adopt Khushi.
A few years later, Madhav, Lucky, and Laxman collaborate with Vasooli Bhai, while Gopal and Laxman work with Babli Bhai. After hearing of Jamnadas' passing, the group comes back together. After 25 years, they go back to the orphanage where they run into Col. Chauhan, Anna, and Pappi, another orphan who frequently experiences memory loss. They discover Khushi's passing when they meet Damini, a woman they mistake for the bungalow's caretaker. Businessman Vasu Reddy discloses the orphanage will be destroyed and moved in order to construct his own structure during a gathering following Jamnadas' funeral. A spirit takes control of Laxman when the party leaves the orphanage and comes back, speaking with Nana Patekar's voice and frightening Gopal. The ghost then takes over Madhav, compelling Vasooli to assign them a task and, inadvertently, returning the three to Ooty. Gopal, who is terrified, dials Anna, who informs him and Laxman that their home is haunted and advises them to move to Col. Chauhan's residence.
As soon as they arrive in Ooty, the two parties begin fighting. The three attempt to frighten Gopal out of the house, but Gopal becomes possessed and severely beats Lucky and Laxman. Vasu Reddy later assigns Madhav, Lucky, and Laxman to set the home on fire. But when Lucky becomes possessed by the ghost and begins speaking, which frightens the group, things start to go wrong. After that, the two groups plan to part ways, but Anna and Damini persuade them to wait for the special occasion instead, and the group mends fences. Gopal has fallen in love with Damini, and the other three coerce him into confessing his feelings.
When Pappi comes, he discloses that the person they believed to be Damini is rather Khushi, who is deceased, and that Pappi's girlfriend, Damini, is the genuine Damini. After Damini admits it herself, the boys become frightened and attempt to flee the house with Pappi, Babli Bhai, and Vasooli Bhai. They are stopped by Anna, who tells that the ghost is actually their long-time friend Khushi.
Returning to the present, Gopal beats up the assailants dispatched by Vasu Reddy as the other boys, who had intended to avoid the situation, are persuaded psychologically to remain. When Nikhil arrives before they can, he says there isn't enough evidence to convict Vasu, which they almost succeed in doing after convincing Gopal to stop following the thugs. This causes a battle, in which Vasu persuades Nikhil to confess while Khushi assaults and almost kills him. Anna then calms Khushi down. Having saved the orphanage, Khushi's ghost bids the boys and Anna farewell before ascending to heaven.
Even though Golmaal Again clashed with Aamir Khan's Secret Superstar, it did exceptional business at the box office, earning close to 315 crore rupees during its entire run and was declared a box office hit. The movie received mixed reviews from critics, with some praising its humour and performances by the star cast but also criticising it for its flaws in technical aspects.
The fifth instalment of the Golmaal franchise titled Golmaal 5 is set to release in the year 2023. Ajay Devgn is reported to be cast once again as the lead actor.
